I'm preparing niosome encapsulating a certain natural extract how can I caclulate the encapsulated dose which is equivalent to the free dose of that substance?
You can use dialysis or gel filteration methods to separate your niosome and free agent then use spectrophotometery.
Also you can use hplc.

You also can use a centrifugation method to separate the niosome from free drug. Then, you can lysis your niosome (direct method) or you can quantify the free drug in supernatant (indirect method).
It is difficult to correlate the encapsulation dose with free dose of natural extract containing many active substances since the nature of active substances affecting their encapsulating ability. The situation is more complicated if there is interactions between active substance on the pharmacological effect. I will select 2-4 active substances containing in the extract as markers and observe their encapsulation efficiency and capacity by Raditya and Alireza suggestion.
