Hi, I'm running some immunofluorescence next week and wanted to sterilize my cover slips in 24 well plates in advance. Does anyone know if I should leave the plates open under uv light or I can close the plates. The protocol I watched online had gheir plates closed, but then I keep some articles say UV cannot penetrate plastic. Plase can someone help? Plates open or not open under uv light?
Already did a trial run this week where I left the plates open, but I want to uv sterilize a lot of plates so please help. Thank you.
Uv can not penetrate plastic. What conditions are enought safe to sterelize your material by open way?? It is the more lmportant issue.

It will depend on the type of UV light that you use for sterilization, but most bulbs made for that purpose generate UV-C light which is not capable of penetrating most materials, including acrylic plastics.
I've always sterilized my coverslips using 70% EtOH overnight in the biosafety cabinet, ensuring they are fully dried before use.

You can put the coverslips inside a clean and dry glass bottle and then autoclave it. That will work. Well, openly Keeping the coverslips inside the hood and allowing UV light to strike, the process works too. So, if you can manage then use a large tray and keep the coverslips on it & do UV sterilization. Otherwise, as John Suggested, 70% ethanol wash.
