PMA and/or PHA do not show any significant effect on adhesion of Jurkat cells. In Jurkat cells, PMA activates the protein kinase C (PKC) pathway in the cytoplasm, leading to downstream signaling through the MAPK/ERK pathway in the nucleus. This promotes the expression of adhesion molecules like integrins on the cell surface, causing weak adhesion to the culture plate. Meanwhile, PHA binds to the T-cell receptor (TCR) on the cell membrane, activating pathways including calcium signaling, NF-κB, and MAPK cascades in the cytoplasm and nucleus, which primarily stimulate T-cell activation and proliferation. Together, PMA and PHA activate pathways associated with T-cell signaling and activation, leading to partial and transient adhesion without converting Jurkat cells into fully adherent cells.
