I'm working on MIC determination using cell culture coated 96 well plate (NUNC Catalogue no. 167008) and just wonder if this will make any difference with using normal untreated plate? I'm dealing with E. coli and S. aureus which don't involve any specific adhesion, biofilm formation and whatnot.
In our experience there is no difference when using treated oruntreated 96 well plate for MIC assay. As an advise, we use plates with round-bottom wells when working with S.aureus: if there is the growth, the cells concentrate in the center of the well and are visible.
For MIC determination, it is okay to use untreated plates, as you do not need bacterial attachment to the surfaces. I agree with Dr Airat - you should use round-bottom wells (or V-shaped wells) to allow the bacteria to concentrate in the center of the well.
I believe that at least EUCAST standards say you should use untreated plates, but I'm not completely sure. Anyway, for drugs like the polymyxins, (and probably for other drugs with high protein binding) untreated plates should be used to ensure correct concentrations of drug in the solution.
