p53 is considered as the Guardian of the cell and suppresses tumorigenic genomic rearrangements. I want to learn more that the exact role of p53 in repairing faulty DNA repair mechanism in vivo following transient cell cycle arrest. Any specific sources/articles/papers and materials on this topic would be much appreciated.
It can activate DNA repair proteins when DNA has
sustained damage.
• Phosphorylation allows p53 to bind to DNA and
activate transcription of cell cycle inhibitors or
proapoptotic proteins.
• It can arrest growth by holding the cell cycle at
the G1/S for DNA damage recognition; thus DNA
repair proteins will have time to fix the damage.
• If repair is unsuccessful then P53 will induce
expression of BAX (acts against bcl2 )and causes
apoptosis. Thus, initiate apoptosis if DNA damage couldn't be repaired
Li-Fraumeni Syndrome is a good example to explain the functions of p53 in preventing DNA-damage to genome that contribute to the development of cancers in individuals. Because p53 has multiple functions (both transcription-dependent and independent), the mechanisms are cell context-dependent and harder to elucidate. However, certain studies have provided insights into the molecular mechanisms.
