Is PC12 cell line always need NGF or It can be cultured with simply RPMI or DMEM with FBS and antibiotic(Peniciline)?
You can maintain these cells in RPMI without problem. However, if you want them to differentiate, NGF is needed. Forskolin (in addition to NGF) can also be used enhance neurite growth. So basically, grow them without NGF in routine and add NGF to differentiate (24? 48? hours ) before your experiment if you want neuron-like cells.

Actually I want to use these cell line for the transaction for the promoter function analysis of Stathmin-1 gene by Luciferase assay. Stathmin-1 gene which is isolated from Mouse brain tissue.
Then it is required to differentiate the cell line with NGF/Forskolin.

We lastly used Bd™ 7s NGF, mouse natural cat.N 354009. 1 Flacon 100 mkg were dissolved in media with 1% FBS is 100x stock.
I culture PC12 cells in DMEM with 10% Horse serum and 5% FBS. I use these cells for Neurite outgrowth, Migration and Proliferation assays to check the activity of Chimeric proteins I developed in my lab
