I made a D64V mutation on integrase for creating a integrase deficient lentivirus.Three day post-transduction, I noticed that not many cells were transduced based on the number of fluorescent cells. So, I am wondering if anyone else also find IDLV result in lower titer?

IDLVs show generally a titer 1-Log below that integrating LV.
In routine we obtain a titer between 1E8-1E10 for LV and between 1E7-1E8 for IDLV (293T and ubiquitous promoter such as PGK, CMV....)

Since IDLVs can not integrate into the genome of targets cells, the promoter used for expression of gene of interest as well as the reporter gene will be less active than integrating LV, and the vector DNA is less stable than that of integrating LV. That is why you see much less fluorescent cells.
