Recently I am doing experiment for EBV supernatnat (virus) collection from AGS+ cells using PEI solution. For the experiment I used PEI stored at -80oC.After collection I checked the virus titer using Daudi cells, but the titer was not so strong.
PEI is very stable when stored at -80. So, your transfection should not be a problem if your cells are permissive to PEI. What type of cells are you using?
Viral production is a bit tricky, so I would spend some time to optimize your viral production protocol (transfection, time to harvest, cells in top shape, etc.).
