I am currently transducing my HeLa cells with lentivirus but I just realized that after DAPI staining the transduced cells always display a "Mycoplasma staining" which is not observed in the same cultures before transduction (example from the internet: http://static.abdserotec.com/thumbnails/Mycoplasma.jpg).

Despite that we have just done a PCR of the culture supernantant for Mycoplasma and the result was negative. I am wondering if some residue of virus RNA remain in cell membrane or something like that. Interesting It remained in cells even after 2 weeks of culture medium change (!).

Any similar experience?
