Does 5% glycerol added to protein buffer (PBS, pH 7.2) interfere with following NMR studies? We are planning to perform NMR based line-broadening experiments to determine small molecule's binding affinity to protein. Thanks!
The hydrogens present in glycerol will likely be present in your NMR spectra. However, you should be able to identify which peaks are due to glycerol and be able to track them throughout your study. With any luck they won't overlap with any other peaks you are analyzing. If you are concerned about your protein interacting with the glycerol you could use an internal standard to quantify the amount of glycerol present in your sample based on your NMR spectra.
