If you are using DMSO or isopropanol to dissolve the formazan crystals, these organic solvents might cause precipitation of proteins derived from the supplemented serum leading to the reduction in the sensitivity and accuracy of MTT assay.
However, the use of other solvents such as 5% SDS-buffered DMF would not require media removal but would need long period of time to efficiently dissolve the formazan crystals.
Malcolm Nobre thank you for your answer. Suppose I want to perform MTT of suspension cells. In which I seed cells in media supplemented with 10 % FBS. Now I cant discard media while adding MTT. In that case what should I do?
