we have been sacrificing mice and rats using cardiac puncture. I am curious if anyone knows of publications or has had personal experience relating to potentially altered protein expression in the heart following cardiac puncture. We are interested in inflammation and oxidative stress in the heart. I would assume that histological staining would be problematic as well. If we are particularly interested in the left ventricle, I assume we should be avoiding puncturing the left ventricle?
Any insights would be appreciated.
Is there any reason why you should use cardiac puncture rather than cervical dislocation in order to sacrifice the animals ?
i would say this is a bad idea
If you are interested in the heart itself to "stress" the heart by performing cardiac puncture is unwise
Cervical dislocation is quick, does not damage or stress the heart - the very tissue you want to investigate - and frankly is less stressful to the animal
