I have adapted my Drosophila S2 cells growing in complete Schneider's medium to serum-free media. After a month of passage in serum-free media, the cells continually exhibit decreased growth rate (compared to growth rate in complete schneider's medium). Also, I added Blasticidin in the serum-free media to maintain the selection for transfected cells in the medium.
Serum-free media is toxic to the cells. They start dying even in few hours in serum-free medium. I wonder how you adapted them. Blasticidin should be added in complete medium. Good luck.

Thank you for your response. I have read from the papers of Khandekar et al. (2001) (DOI: 10.1006/prep.2001.1448) , Kleditzch et al. (2003) (DOI: 10.1016/S1046-5928(03)00139-6) that they have successfully adapted Drosophila cells in serum-free media.
