I am trying to perform anti-BMP-2 IHC in formalin-fixed rat femur using Abcam primary antibody (ab14933). The manual suggests spleen, lung and intestine as positive controls but until now I didn't succeed using HIER or PIER protocols that are working with other antibodies. A protocol from University of Rochester indicates the growing plate as a positive control but can't detect anything. Any suggestions for positive control and for whether or not I should suspect the primary antibody.

I did IHC on BMPs 10 years ago (no Abcam at that time). Most BMPs should be detected in steoblast cells, especially those forming new bone, so you do not need other positive controls.
