try Total exosome isolation (from plasma) reagent. There are two protocol options- with proteinase K and without, depending on your downstream assays. Plasma is probably the most challenging sample type for isolation of exosomes/EVs- as its viscous and contain plenty of "contaminants" - misc proteins, aggregates, etc. Ultracentrifugation is still one of the most popular approaches for isolation of exosomes, but it takes a while to master it and its a lengthy and tricky procedure. Thus we are always working on improved, robust, fast, simple and scaleable procedures

I also want to know how to isolation serum exosomes in high yield. By the ultracentrifugation,20ug protein can be get from 100ML serum.It is so poor for my next assay.So if you get some good methods,let me know.Thank u

ultracentrifugation recovers just a small portion of exosomes, but its great for processing large volumes. with reagent, our typical vol for serum is 100 ul - 1 ml, we recover plenty of exosomal material- enough for all typical types of downstream analysis; but its hard to process large vol like 100 ml- as you said
