My knowledge is quite limited in the mRNA translation field, but I can see in the literature that most of the translational mechanisms rely on the presence of the UTR sequences. I was wondering whether there are other alternative mechanisms that regulate the mRNA translation solely depending on the cDNA sequence. That would explain how generally the sequences cloned into vectors are regulated.
Which system are you going to use? If cloning in yeast that may work, however, expressing in bacteria may not work.

in higher eukaryotes also splicing has a strong impact. Unspliced transcripts tend to be translated with low efficiency.
Regarding the vectors, almost all have some kind of regulation sequence, which kind of sequence is depending on the specific vector and/or vector type.
