We've done some tests with an enzyme immobilized in CM5 and NTA chips. We've tested both small molecules and antibodies, and in the majority of the cases the relative units (Sample-Blank) give a negative curve from the association-dissociation process. I know that the Biacore only measures the angle of the light reflection, and in this way the changes that come from the plasmon resonance by the interaction of the ligand and the analyte. That said, I've never seen negative curves anywhere, and even when I can measure a Kd by these negative curves, I don't know if this would be correct. Can anyone with experience in this field tell me if this is normal, or only an artifact? What can I do to check if the results are reliable?
