Hi researchers, I am evaluating the plant extracts against Parkinson's Disease. I want to know which is the common cytotoxic agent to induce cytotoxicity in SH-SY5Y neuroblastoma cells. If someone knows this can they kindly share it? All the efforts will be highly appreciated.
usually in our lab we first differentiate SHSY5Y cells with retinoic acid for 7 days (though some lab also work on the non differentiated cells as well) and use MPP+ or 6-OHDA to induce neurotoxicity relevant to PD. The time and dose of the toxin usually needs to be standardized, though you can get an idea from the papers. this two are the most widely used toxins for developing PD cellular model.
I agree with Priyankar Sanphui, - differentiated cells are more relevant model to PD than non differentiated. Concerning to MPP+ and 6OHDA, they induce (neuro)toxicity via oxidative stress, however, they differ each other in the proper mechanism.
I agree with Priyankar's suggestion. Regarding stressors you can also try paraquat (it is highly water soluble) and a herbicide linked to Parkinson's disease.
