Dear AAV-community,
I am looking for data on excretion kinetics of AAV9 viral vectors from mice.
So, I need the information how long urine and feces contain infectious AAVs after i.v. injection.
For AAV2 the German genetic genetic engineering commitee cited:
Kay, M.A., Manno, C.S., Ragni M.V., Larson P.J., Couto L.B., McClelland A., Glader B., Chew
A.J., Shing J.T., Herzog R.W., Arruda V., Johnson F., Scallan C., Skarsgard E., Flake A.W., High
K.A. (2000). Evidence for gene transfer and expression of a factor IX in haemophilia B patients
treated with an AAV vector. Nature genetics, 24:257 – 261.
for: "In addition, vector sequences are no longer detectable in the blood and urine after 48 hours, meaning the propagation of infectious AAV vector particles is restricted."
I would need this data for other serotypes as I need to handle animals that are excreting GMO viral vectors differently. Although it is common sense that after one week there will be pretty sure no AAV vectors excreted, I need to proof this or have to handle the mice under the more strict conditions for the whole time of the experiment (which is expensive an unnecessary).
Can anyone help me with this?
Thank you a lot!
Volker
