Does anyone know a method for mitochondrial detection with FACS using Mitotracker green (Life Technologies)?

We are currently trying to establish a method for detection of mitochondria using fluorophore mitotracker green. While this dye is suggested to only show in the green channel, it is also appearing in the red. The trouble is, we are trying to dual stain for mitotracker green as well as TMRM, therefore a clean signal from mitotracker green would be ideal. Has anyone else experienced this issue? We have already asked the suppliers for a different batch, but the problem appears to continue. Any suggestions?

Hi Kirti,

I had best results with using Abcam mitochondrial marker mouse monoclonal antibody instead. It's anti-mitochondria [MTC02] antibody (catalog number ab3298). Very selective, and you are not limited by the choice of the fluorophore - pick any of the multitude of the Alexa-conjugated secondaries your heart desires. Excellent and highly specific to this organelle marker. Besides, it won't depend on the mitochondrial potential like most of the charged dies are. I hope it helps. Good luck!
