Aphidicolin prevents mitotic cell division by interfering with the activity of DNA polymerase-alpha. It has worked amazingly in our lab, but we'd like to try a different, less expensive drug that can inhibit DNA synthesis without affecting DNA methylation or RNA, protein and nucleotide biosynthesis.

have you checked on hydroxyurea? We have used it in the past to inhibit cell cycling (on plants). Many other labs use this: e.g.
Mol Biochem Parasitol. 1996 Sep;80(1):89-102.

But please check on side effects.
