I am reviewing relevant literature for DNA extraction from this bacterium. Considering that my objective is cloning, which is the best protocol to be followed? Which kind of culturing techniques, mechanical lysis or not, etc. Thanks in advance
u may try MP Biomedicals soil DNA extraction for pure culture DNA isolation subject to availability in your region, as it works best in terms of RNA and protein removal, we use the same for genome sequencing.
I was in Molecular Diagnostics Laboratory for M.Tuberculosis and there we use to Isolate the DNA from Clinical Samples... for this firstly we use go for Decontamination prodecure by NALC-NaOH (modified Petroff's Method) and after that we use to isolate the DNA by Axygen Kit....
In the past when I was clonning from TB, I found it easier just to order up the DNA from the ATCC (http://www.lgcstandards-atcc.org) or another or european equivalent, saves a lot of trouble if you dont have BCL3 facilities.
