Hi all, a question for the protein purification masters here. After protein elution from a Ni column, some people simply dilute the sample down to decrease the imidazole concentration. I have always done buffer exchange (4L, O/N, 4C) and feel this is the best approach to get rid of the imidazole. Has anyone ever used a simple dilution step? I dont think this is the best approach, but it would significantly save me a lot of time.
