I have tried to design Tough Decoy inhibitor for miRNA 195, so I want to synthesize my tough decoy sequence on the downstream of a eGFP gene but unfortunately I do not know how much distance should I put between eGFP and tud !!

Do you think it is ok putting the TUD directly after eGFP ?

How many Base paires should i put between eGFP and tud will makes this tough decoy more effective?
