I'm trying to a kinetic methyltransferase assay and would like to use ziptips as a means for quick RPLC, but I'll need hundreds of ziptips to generate good data and it just seems like a really expensive venture to buy them. I'm wondering if anyone out there has a protocol to take regular c18 or c4 resin and pack their own ziptips in micropipette tips. Any help is much appreciated! Thanks!

You get C18 in a paper form. Take those syringe needle which is used to load samples in hplc. Take such syringe needle which has a bit wide opening (sorry right now i am not able to recollect name of this syringe). Put that needle on the syringe and poke holes in that C18 paper. By doing this you are creating C18 paper disc in that needle. After sufficient pokes, insert the syringe in the tip like pipette. And now inject air. This is displace those C18 paper disc from needle to the tip.
That's it. Your own ziptip is ready ! ! !

The best source is by 3M Empore Disks. They are sold as C18 extraction disks. The protocol is originally from Mann Lab. Here is the reference:
Protocol for micro-purification, enrichment, pre-fractionation and storage of peptides for proteomics using StageTips. Nat Protoc 2, 1896-1906, 2007.
Using the yellow pipette tips, you can pack hundreds of these disks and have a good supply that should last you for a long time.
