you could try use specific media for actinomycetes isolation such as starch-casein agar (SCA), humic acid-vitamin agar (HVA) or SM3. Please find below the references and target genera for these media:

SCA (Streptomyces and other actinomycetes) - Küster, E., Williams, S.T. 1964. Selection of media for isolation of streptomycetes. Nature 202:928–929.

HVA (rare Streptomyces) - Hayakawa, M., Nonomura, H. 1989. Humic acid-vitamin agar, a new medium for the selective isolation of soil actinomycetes. J. Ferment. Technol. 65:501-509.

SM3 (Amycolatopsis and related genera) - Tan, G.Y.A., Robinson, S., Lacey, E., et al. 2006. Exploration of Amycolatopsis diversity in soil using genus-specific primers and novel selective media. Syst. Appl. Microbiol. 29:557–569.

i ussually use Mannitol Soya Agar, but other bacteria also growth there. even though I was just expecting Actinobacteria that grows there hahaha

Hi Gema,

Am also working in the Actinomycetes field....Here am providing the protocol for you to isolate actinomycetes from soil sediments.

1.Actinomycetes from the soil had been isolated by pour plate technique Actinomycetes Isolation Agar (AIA), also Supplemented with Actidione 20 mg/l and Nalidixic acid 100 mg/l were added to minimize the other fungal and bacterial growth respectively. Soil samples were serially diluted and 0.1 ml of aliquots were spread over actinomycetes isolation agar plates. The plates were incubated at 28±2 °C for 7 days. Strains of actinomycetes were picked out and purified by repeated streaking on Yeast extract-malt extract agar (ISP2) and were preserved in slants at 4±2 °C.

2.Suggestions:

Instead of the Actinomycetes isolation agar you can try with Starch casein agar...Otherwise both you can try. You will get good actinomycetes cultures.

You could try Gauze 1 mineral agar (Gauze et al., 1983) - I use it for isolation of soil actinomycetes (the best for them). But also you can try R2A.

and do not forget to use some antifungal agents (e.g. nistatine or cycloheximide)

Make your media by trying the use of antifungal and antibacterial combinations including pre-heat treatment of soil samples. Still there is no single medium which can selectively isolate all actinomycetes.

Pre-treat the soil sample as follows

Dry the soil at 45C overnight

Add the 1N NaOH and 1: 10 CaCO3

again dry at the 45C temperature and

Prepared the soil suspension followed enrichment for 248 hours in actinomycetes selective broth and by streak on same soild media

You can also add the more agar agar powder to provide the further selectivity to the actinomycetes species

Hi Gema, dalam bahasa Indonesia ya. Pengalaman saya waktu S2 , isolasi aktinomisetes menggunakan media chitin agar dan aktinomisetes terutama streptomyces banyak tumbuh pada medium tersebut. Disamping itu medium water agar juga dapat digunakan, karena aktinomisetes dapat tumbuh pada medium yang sangat miskin. Bisa juga secara sederhana menggunakan media ekstrak tanah. Semoga bermanfaat.
