Suggested Bates et al. protocol is still the best protocol for determing proline levels. Our research team is first to demonstrate accumulation of proline in plants exposed to heavy metal stress (Alia & Pardha Saradhi, 1991 - you may download the same from my ReserachGate account). You may download several other papers we published on proline.
I do not recommend centrifugation. You can withdraw toluene layer containing colured proline-ninhydrine complex carefully with a dry pipette. If you wish you may visit my lab at University of Delhi, as my scholars use this method frequently and you can directly have a practical experience.
I think it should be much easier for your lab to make a short film (it can be even accomplished by using a iPhone) and put it on YouTube, so other scholars from different parts of the world can just learn it from the film without traveling the long way to visit a lab. Nowadays, a lot of Universities, companies, scholars are taking advantage of YouTube for teaching and other purposes. Just a thought. Best.
