Hello. I'm interested in transfection of Panc-1, MIA PaCa-2 and Capan-2 cell lines with microRNAs (transient transfection). I know these pancreatic cancer cell lines are difficult to transfect. So, does anyone have any experience with transfection with these cell lines, and an available protocol for microRNA transfection with Lipofectamine 3000 or Lipofectamine RNAiMAX reagents for them? Or any tips?

miRNA transfection is pretty similar to siRNA transfection (in terms of the mechanics and cell stress) so you would be well off using similar protocols. For tough pancreatic cancer cell lines, you might want to look into specialized reagents (e.g. for PANC-1: https://altogen.com/product/panc-1-transfection-reagent-non-endocrine-pancreatic-cancer/) as these are likely to have higher transfection efficiencies.
