My protein has a PI in the physiological pH and is in the phosphate buffer, pH 6.0. I would like to check if the protein is in the monomeric state.

Blue native PAGE, Nat Protoc. 2006;1(1):418-28. It works fine, even with hydrophobic membrane proteins.

By the way, size exclusion chromatography (e.g. Superdex 75 or 200) coupled with a Light scattering detector is a better option to check the monomeric state.
