We have been synthesizing nano-particles biologically in my lab. We are thinking to use them as delivery carriers. We have been advised to perform immunological assays,before using them, to study whether any immune responses are generated from them. We are afraid that proteins binding onto the particles would arise any immune responses that might be adverse. Since our lab is more concerned about bacteria and yeast fermentation, and we are very new to this field, we are not aware of any immunological studies. We would be more glad to know, how the immune system responses could be studied when the nano-particles are administered.
It's very hard to answer to this question, because you have to really focus in one deal, or take it step by step, you have to know some details about the imune system, first of all, what do you want to know exactelly??? if you want to just know the toxicity of the immune system caused by your nanoparticles, that's a deal, if you want to know the effects of those nanoparticles on bacteries and yeasts anti-microbian activity or else), that's another deal, but if you want to use your nanoparticles in drug delivery this is really another different, extremely different, deal, first of all you have to know which route of administration that you will apply, after that, you have to know, which kind of bioassays that you will do, in vivo or in vitro, which kind of organisms if you will do in vivo assays, which kind of cells in case of in vitro assays, how your nanoparticles interact with your organisms, cells??? of course you have to know the physical and chemical properties of you nanoparticles, the surface charge, the composition of the surface, size and shape of those particles???? what would you exactelly know??? cell viability, oxidative stress, iflammatory profile, genotoxicity, protein expressions, specific protein expression or what??? i think if you try to answer to those few questions you can find you road, Good work,.
NPs are predicted to induce innate immune responses directly and indirectly. Therefore, follow the literature. There are number of published studies.

Happy to know you and thanks for letting to know more. I am going to elaborate little more about the nano-particles and their future progress, we are planning to do. The size of the particles is 50-150nm, which is spherical and it is constituted by lipids (phosphatidycholine, phosphatidylserine and phosphatidylinositol). Since, the particles are derived biologically, usually the particles surface is filled with the proteins. We do have the proteome of the particles. So far, we didn't studied the surface charge yet. The objective would be to deliver the exogenous therapeutic substances (proteins, siRNAs and chemotherapeutic drugs) using these particles in mammalian cell lines, which we didn't selected yet. We are aiming to follow both passive and active targeting studies at in vivo and in vitro conditions. No perspective of using the particles to bacterias and yeasts. As you stated, the first phase of our studies would be the toxicity of the immune system caused by the lipid nanoparticles, without any therapeutics. Once the latter studies are well-defined and analyzed, we might get green pass to go forward in using them as delivery carriers. We are taught to administer these particles in syngeniec and allogeniec mouse, which in case would provide the immune toxicity responses. i hope, i have presented my views to my knowledge. I would be glad to know, how administration routes influences the escape of particle from immune system?.

I am biased toward neutrophils (PMNs) since that is what I have studied most of the last 18 years. I would want to know if isolated PMNs phagocytose the NPs and if an oxidative burst is initiated. While this is not a toxicity response it would tell if they are immunoreactive.
