I would like to go for mode of action studies for bio active compounds from plant source, for that it will be helpful if i get some studies involving methods other than, identification of membrane disruption against bacterial test pathogens.
Please, find in attach an article on: Antibiotic development challenges :the various mechanisms of action of antimicrobial peptides and of bacterial resistance

You can try to do macromolecular labeling to determine whether your compound hits any of the macromolecule biosynthesis, such as DNA, RNA, protein, cell wall, and lipid pathways. It's based on incorporation of radiolabeled precursors. Expression profiling can also be performed and compared to that of drugs with known mechanism of action. Isolation fo resistant mutants can also identify a direct target.
Antimicrobial activity by change in membrane permeability is only one of the methods. There are other possibilities like inhibition of protein synthesis, inhibition of DNA synthesis, inhibition of many catabolic pathways etc.
