I'm looking for a way to dissolve my dried antibody which was aliquoted and stored in -20 C for at least one year. Right now I need to use these antibody for western but somehow it got dried.
I think the anti freezer glycerol may have evaporated.
But I need to use it so is there any way to re-suspend the dried antibody.

Hi Banshi, I agree with Caroline in that 50% buffered glycerol is probably the best way to go. Abs are usually very stable proteins and they can take a lot of abuse.
Polyclonals are usually more stable than monoclonals.
Caroline, I have also seen this so many times and I will also have to yell. DO NOT USE FROST-FREE FREEZERS TO STORE BIOLOGICAL MATERIALS.
I actually haven't had problems storing antibodies or enzymes for years in our lab's frost-free freezer (the PI chose frost-free for ease of maintenance). As a precaution for those of us who have to use such freezers, you can use a stratacooler that will resist changes in temperature during the defrost cycle to hold important reagents.
Hi Banshi, You can resuspend the antibody using 50%PBS and 50% glycerol. But I am not sure that you can detect your protein of interest except a lot of background and non specific detection. After re-suspending the antibody if it seems yellow other than transperent i.e you can not use this antibody because it has been oxidized.
I don't know how your antibody could evaporate. If you had tightly closed the vial is impossible that glycerol can be evaporated. Anyway, you could add the same buffer that you used when the antibody was freezed. You have to take into account that the salts content will be twice that in beginning. If you remember the volume in the vial, you may add only water and the salts content will not change.
