I have to stain the gonad of the C. elegans (outside the worm). For the same I need it to come out intact without breaking anywhere from the distal to proximal end.
Transfere the worms to a drop of buffer on a glass slide. Use a thin syring to cut off the head just behind the pharynx or cut off the tail. Then the gonad arms will put out together with the intestine. If you cut it to low you will also cut the germline.
It is helpful and I am doing the same thing. My concern was to after they get out how to keep them intact for staining. I usually freeze them on slide with a cover-slip on it and then flip the cover-slip for further protocol. I hope there is any other way to keep them intact on the slide without being getting break ......!!!
After the gonad went out you can add 8µL of PFA, put a cover slide, pour the slide in liquid nitrogen and remove quickly your cover slide with your finger. (freeze cracking)
