We would like to silence BuChE in HepG2 cells, but we have a problem with the BuChE occuring naturally in serum. In some lab soman was used to inhibite AChE and BuChE in serum. Is it possible to have some serum with soman ( or just soman-treated and then soman is somehow cleared away?), or some serum without BuChE and AChE?
I know from CHO cells that they can be cultured in completely protein-free cell culture medium. Under these conditions they form sphaeroblasts and must be cultured in roller bottles. To prevent foam formation you must add some antifoam reagents. Maybe this also works for HepG2 (I found this reference: http://www.ncbi.nlm.nih.gov/pubmed/9548366). In the case of CHO it took a longer period (3-4 weeks) to adapt the cells to the protein-free medium and at first a lot of cells died. After this period the survivors started to grow and proliferate again and could be stably maintained in culture.
