I would like to measure the concentration of NO in rat tissues. Is Griess reagent ok? It measures only NO2. Can I measure it also in samples from frozen and unfrozen tissues (not fresh)?
I have tried Greiss reagent, DAF staining, and NOanalyzer before. Yes, you may try any one of it but just be aware to avoid nitrite/nitrate contamination when you prepare your working solution. I think Greiss reagent measures NO2 (nitrous oxide) is a mistake you must mean NO2- (nitrite). You can acutually convert NO3- into NO2- using cadmium method or E. coli nitrate reductase to covert NO2-/NO3- into all NO2- and measure it. Good luck.
Thank you for your answers. Of course, I mentioned NO2-, not NO2, - is missed somewhere. :-). Does this mean, if I would measure NO2- only with Promega kit, I would bring me no usable result? Is there no ratio between NO and NO2- and between NO3- and NO2-?
