I’ve been trying to generate IFN-gamma secreting NK cells from murine progenitors (Lineage- Sca1+ cKit+).
I’ve tried various protocols with differents combinations of cytokines (including human FLT3L, murine SCF, human IL-2, human IL-7, murine IL-12, murine IL-15, murine IL-18) with differents stroma (OP-9 cells, MS-5 cells) and succeed into generating a large proportion of CD3- NK1.1+ cells (up to 80%) but so far these cells don’t produce IFN-gamma ; neither spontaneously nor upon stimulation (IL2, PMA, IL12+18, LPS…). Have anybody succeed in this field ?

Thank you for your answer Sharafaldin, however this paper is my bedside book for quite a while and up to now i did not find anything practical in it susceptible to solve my problem...did you ?
I'm not sure about murine cells but IL21 might lead to another differentiation state.
I think triggering of NKp46 could also do the trick.
Have you tested them for cytotoxicity?
