Our 786-O xenograft is done with 1 million cell injections (http://altogenlabs.com/xenograft-models/kidney-cancer-xenograft/786o-xenograft-model/). Usually the problem I see with other protocols is that they don't use confluent cells, and the problem there is that you might not get stable tumor formation. Of course you need to use immunodeficient mice, otherwise you might get an immune response that kills the tumor.

I dont know if you used matrigel or not but I would suggest use of matrigel if you have not used it. Alternatively I would use a more immunodeficient mouse model like NOD/scid or NSG.

I am unfamiliar with the cell line you are using, but it sounds like NK [natural killer] mediated rejection. NOD/scid and NSG mice have lower NK activity due to the effect of NOD [non-obese diabetes]. If you prefer a nude-based model for ease of tumor tracking, I would suggest bnx [beige/nude/xid] mice, as the beige mutation also inhibits NK activity. A final method for blocking NK activity is treating the mice with anti-asialo-GM1 antibody [commercially available from Dako], but you would have to dose the animals chronically [every other day, I think], which is not cheap.

I'm not familiar with CD1 Nude, but very familiar with Nude or SCID on Balb/c or C57Bl/6 background. Sounds like your mice were not really T cell deficient. If you reorder, use SCID mice (T and B cell deficient). Because tumor grew and was rejected, it was due to T cells, not NK cells.

yes CD1 NUDE is not completely immunodeficient like SCID/NOD.i konw CD1 nude have precursor of immature T cells live...that could be one reason, but i had tried before with other carcinoma cell lines(like HCT116,A2780,SKOV-3...) and observed proper tumor growth.why it is so??

My suggestion is use and try different amounts of cell like instead of 5 million increase the amount try10, 20 million of 786-0(RCC) cells and see try in all three immunodeficient mice like Nude, or Scid or NSG and give more time to grow the Tumor. Good luck

So far I am not working on that model now a days. If you are facing difficulties in tumor take up and growth  then I feel you should go for SCID/NOD SCID mouse models for 786-O xenograft.

The routine for renal adenocarcinoma/786 is the fast growth followed by disappearance of tumor due to NK response in nude mice. In a pilot that I conducted with three animals  I had successonly in one animal, but after re-inoculation of 4X10E6 cells. The literature reports that the xenograft works in BALB/c nu beige or SCID but I have no experience with these strains.

I have the same problem, I'm using the Swiss Nude, I had inoculated 10^7 cells 786-0/animal (cells resuspended in 200 µl of medium serum free)

After one week I observed tumors and it disappeared after the second week

I hope you solved your problem. What could you suggest for me ,,,,???

Our lab have been able to grow 786-0 subcutaneously flank tumors in scid-bg mice with an inoculum of 5 million cells in 50% matrigel.  The source of the cells is ATCC.  Can size match a trial in about 35 days (225 mm3).
