Following DNA damage and senescence induction it can take up to 7 days (Coppe et al, 2008) for the SASP to fully develop. Since it has been suggested (Burton et al 2010) and evidence provided (Liu et al, 2013) that the SASP may play a role in senescence mediated calcification, we may not be giving enough time for the calcificatory phenotype to develop.

The onset of SASP in cells following DOX treatment may take longer time. Further, the phenotype onset is likely to be asynchronous. Therefore, to make sure that cells are exhibiting SASP, levels of proinflammatory cytokines need to be measured every other day. After establishing SASP in cells other phenotype can be followed.
