I am not really sure what "new systems" really means.
If you already know that your protein of interest may be internalized via CCVs, you may co-stain your cells with genuine CCV markers (i.e. CHC, etc.) and your protein and simply evaluate colocalization. Be aware that CCPs are below the diffraction limit.
If you want to discover new cargoes, there are only two possible approaches:
- candidate approach implying that you test whether protein X or Y is in the CCPs?CCVs
- unbiased approach requiring the purification of CCVs (see for example Galovic et al., JCS 2011) and subsequent identification of their content by MS.
thanks for the ansewer...I'm already costainiing my protein and CHC and I was thinking to isolate CCV, which is not easy as far as I know that's why I was searching for new system!
