I am trying to measure the permeability of Human Brain Microvascular Endothelial Cells against certain compounds with TEER, functional assay with FITC-Dextran and checking the expressing of Ve-cadherin (Immunocytochemistry). My concern is should the cells have to be serum-starved for measuring permeability?

Serum-starvation usually gives you higher TEER values (and lower permeability) due to induced tight junction protein expression. If you want to study the direct effects of the compunds (and compounds do not bind to serum proteins), I would suggest to study the effect without serum-starvation!
