Im trying to setup an NK-assay using NK-92 as effectors and K562 as targets.

I use a 96-well round bottom setup with 5x104 targets and 1:1 till 80:1 ratio E:T.

After 2-4 hours i measure LDH release.

As part of the controls I have wells with just NK-92 cells and no K562 cells in the same amounts and medium as in the E:T wells.

My problem is that, these control wells show an increasing amount of LDH release suggesting the NK-92 cells autolyse (or are killing other NK-92 cells). This occurs in wells +IL-2 as well as in wells without IL-2.

What could be going on here? and how can this be prevented?
