I have come across many studies regarding isolation of monocytes from PBMCs and then culturing these adherent cells. To differentiate the monocytes to dendritic cells I understand that GM-CSF and IL4/3 is required (TNFalpha for mature dendritic cells). For monocyte-derived macrophages I have come across publications which states that the addition of M-CSF to the medium is required. However, there is also some suggestion that incubation with serum is enough for macrophages to differentiate after 5-7days. Is this correct? If I incubate my isolated monocytes with serum (no addition of M-CSF) for a week, what will I actually have? Any help or advice would be greatly appreciated.
