I am trying to culture cancer cells in microfluidic device, but after 72 hours, the device starts to look fuzzy under the microscope. What is causing this fuzziness?
If you use PDMS for the fabrication, it has nothing to do with the cells. I've seen people culturing cells and tissues in PDMS for a week or more.
And what exactly can you see under the microscope? Do you see any deformation in your channels? If not, I believe there might be a contamination causing the fuzziness. Sterilizing microfluidic devices is a bit tricky
If you are using PDMS on your device, depending on the hardening temperature you used, the PDMS may be very porous. Therefore, some diffusion may be happening and, consequently, causing the fuzziness.
As Dina said, also check for contamination. Due to the microstructures, microfluidic devices are sometimes hard to sterilize properly.
