I want to make a DIY enzyme using simple methods but wants to be more scientific. Using yeasts, can they take up and reproduce enzymes from fruits juice culture? If there are different enzymes in the culture, will each yeast cell take up one enzyme at a time or possibly more than one enzyme?
What you are proposing is not how enzyme production works.
Enzymes are usually proteins (very occasionally RNAs). There is no known method by which an organism, for instance a yeast or bacteria cell, can use a protein as a template for production of more of the same protein (prions are the only thing that comes close). Enzyme production usually occurs by an organism producing a protein that is encoded within it's own DNA.
Making an enzyme artificially is usually achieved by giving an organism an artificial DNA that contains the instructions to make the enzyme, usually a DNA plasmid. The organism can then make the enzyme, and the enzyme can then be purified via several methods (e.g. affinity chromatography).
The cheapest and easiest path to DIY production of a specific enzyme would probably be to transform a plasmid from Addgene encoding the enzyme along with a Hexahistidine tag into a suitable strain of E. coli, such as BL21, then culture them, collect them, lyse them and crudely purify the enzyme by NiNTA.
Even that simple procedure would require quite a bit of lab infrastructure, however (incubators, centrifuges, etc.). If you don't need to purify the enzyme you could simplify the production a bit. What sort of enzyme did you want to produce?

If you're interested in obtaining a certain activity, e.g. enzymes that can digest certain carbohydrates or proteins, a common way is to grow a fungus on the material you want to digest, forcing it to secrete appropriate digestive enzymes. There are a lot of articles you can find easily online that characterise fungal secretomes produced in this way.
In the blog-post you link to, they are using unpurified mixtures of proteins, and that's probably good enough for your purposes. Otherwise, you're going to have to look into protein purification, and use the recombinant techniques described by Brian in his response.
