I am going to run a time-course experiment for CHO cells, and the cells will be grown in 250 ml glass (instead of polycarbonate) shake flasks. However, I have some concerns that glass may not be suitable for CHO cells and could affect their density and viability. Do you have any idea about this?
If you're trying to maintain a suspension culture, glass vs. plastic shouldn't have much of an effect, as the cells will not interact much with the walls of the flask. Just ensure you have appropriate controls.
However, if you're trying to get the cells to adhere, then you will likely need to coat the glass to foster good adhesion and promote growth, for example using fibronectin. At the very least you will want to treat the glass with NaOH to make it more hydrophillic.
