I have found that in most papers Ca2+ and Mg2+ free HBSS was used to prepare B16-F10 cells suspension for the in vivo metastasis assay. I performed a similar assay recently, but unfortunately used HBSS with Ca2+ and Mg2+ by mistake and the result was rather poor. I have been wondering if the poor result might have been caused by the Ca2+ and Mg2+?

I have been looking for references to explain this but have not yet found any. However, I have found an article [1] that says that Mg2+ supports B16 cell migration in vitro and also that Ca2+ does this under some conditions.

I am also going to perform in vivo LLC (mouse Lewis Lung Cancer) metastasis assay. What effects do you think Ca2+ and Mg2+ might have in this case?
