Dear fellows,

I am perplexed with some DNA damage response in my osteosrrcoma cell lines. I am treating the cells with the same concentrations of three cytotoxic agents: a novel melphalan derivative (MPH-d), melphalan (MPH) and doxorubicin (DXR). All three reagents induce T68 phosphorylation in CHK2, but only the melphlan-derivative is capable of inducing phosphorylation of H2AX and apoptosis. Could you please help me with the interpretation? Are H2AX and CHK2 phosphoryation independent events or inter-dependent? Thank you in advance.
