I am going to start my protocols in RNA isolation, both qualitative and quantitative. What difficulties should I have in my mind? and what will be the solution for that?

For good RNA yeild you should try and do most of your work on ice. Also make sure that you use RNAse/DNAse free water and keep RNA frozen (concentrated). Only dilute small amounts when required. We use Qiagen kits for RNA extraction- very easy very convenient.
I have been trying to extract RNA from wheat leaves using Trizol (invitrogen), but sometimes fail to get clear cut bands, There is some inconsistency in the efficiency of the method. Can anyone suggest any modification of the same to improve the efficiency? Thanks a lot
