I want to assess IL-1alpha in cell culture supernatants with ELISA. There are a number of suppliers selling kits for this purpose and also pre-coated plates, with a huge variation in prices. My question is, does anyone know whether the difference in price also reflects a difference in quality? Do I gain a lot of time and get better results if I go for the more expensive kits, when the application is as "simple" as cell culture supernatant (without serum)?
I also wonder what reagents I might as well mix myself and what reagents are developed and optimised by the manufacturers? (wash buffer, reagent diluent, stop solution etc). I want to avoid lengthy development and validation work, but at the same time I do not want to buy e.g. a really expensive "stop solution" when simple 2N sulfuric acid from the lab next door will do.
