Does anyone know a method/stain that I could use to identify transdifferentiated cells that were previously fibroblasts. The most robust method would be maybe to use a transgenic animal using the a Cre/LacZ cross with periostin driving Cre expression, but Im wondering is there another way without generating this model. Specifically ISH/FISH approaches?
I welcome any suggestions.
You could try labelling of fibroblasts with BrdU. My limited experience with this technique indicates that the cells were not as healthy looking. Others with more experience may be able to comment on this.
Alternatively implanting cells derived from a male donor into a female host could be used. Am currently looking into best way to do this myself.
Implanting human fibroblasts (mesenchymal stream cells) into an animal host tissue could also be used with the human cells being tracked using an antibody to human nuclear antigen. Am in process of testing this right now. The cells seem to have been well tolerated but have yet to do histology.
